Promoter-GAL4 Vectors
A restricted ectopic expression pattern can be achieved by fusion of a vector containing the GAL4 gene and hsp70 terminator (pGaTB) to a cell type specific promoter fragment. The construct can then ligated into a P element vector such as pCaSpeR4 for integration into the Drosophila genome.

Alternatively a new vector from Sharma and colleagues (2002) is available for quicker construction of your promoter-GAL4 vector:

A restricted ectopic expression pattern can be achieved by cloning a specific promoter fragment into pPTGAL (map , sequence, reference)

UAS-gene constructs

Ectopically expressed GAL4 will activate transcription of a reporter gene or another target gene that is downstream of a UAS site. These constructs are made by ligating the gene of interest into the pUAST vector:

pUAST (map , sequence , restriction map). 

Please note: this pUAST sequence is now correct to our knowledge - BUT please sequence when making fusion constructs that have reading frames running through the MCS

pUAST-NDam (sequence) - for DamID chromatin profiling experiments (Dam-myc cloned into EcoRI/BglII sites)
